Process Analytical Systems

Abstract:

The ability to characterize input materials and monitor a process in real time enables sophisticated control strategies that maintain tight quality specifications regardless of perturbations. The presentation will discuss the engineering tools and knowledge required to design and deploy Process Analytical Technology (PAT) solutions in pharmaceutical drug substance and drug product manufacturing as an example of advanced sensing techniques which enable process control. Current PAT technologies will be discussed including the engineering theory, principles, and mathematics required to design and deploy these technologies in a pharmaceutical manufacturing environment in compliance with FDA and international regulations. There will be an overview of analyzer types and principles of operation, chemometric techniques for multivariate analysis, multivariate process models, dynamic process control, and advanced pattern recognition techniques. In addition, the presentation will cover the technical aspects of real-time data management and 21 CFR Part 11 compliance.

April 9: Efforts in sustainability -

a discussions of tools

and cooperative initiatives across industries

Abstract:

Many large retailers and brands are looking to improve the sustainability performance of their suppliers (where the majority of the life cycle impacts of the products they sell actually occur). However, due to the lack of direct control over the methods of production, retailers and brands are using targeted questionnaires and Indexing to collect relevant data to measure performance and work to motivate improvement in the supply chain over time. Two examples of indexing in the US (in the apparel and consumer goods retail sectors) will be touched on.

Michael will also give an overview of Life Cycle Assessment / Life Cycle Thinking, a driving tool in sustainability, and share how the Business & Institutional Furniture as well as Electronic & Electrical products industries use LCA and Environmental Product Declarations to communicate to B2B or consumers. A connection will also be drawn from these tools to the LEED program and potential cost savings.

About the Speaker:

Michael Richardson, P.E.,LCA & Sustainable Design Sr. Project Manager

Michael obtained his Bachelor’s and Master’s degrees in Chemical Engineering from Cornell University, and has his professional engineer’s license from the state of New Jersey. As a sustainability consultant at SGS Michael supports clients’ sustainability efforts by helping them measure, reduce and communicate their environmental impacts, though Life Cycle Assessment (LCA) and other tools. He also coordinates with the SGS global sustainability team, to facilitate execution internationally when expertise is needed within supply chains. Prior to joining SGS Michael worked in the environment engineering field for over 10 years: modeling contaminant generation; implementing site remediation; as well as conducting LCIs and LCAs, on products ranging from packaging to flooring.

Currently Michael is supporting clients in: the electronics Industry to prepare Environmental Product Declarations; and the cleaning product industry for environmental labeling. He has also been an active member of the Outdoor Industries No-Harm-Down working group and is co-representing SGS on the verification committee for the Sustainable Apparel Coalition.

Abstract:

Effective operation of fluidized bed processors requires even gas distribution and effective gas-particle mixing. Scaling up a drying process from laboratory to production scale presents unique challenges, such as preventing granule attrition, fines loss, or segregation. Current methods make optimizing processor performance difficult without large-scale experimentation due to the scale-dependent parameters that affect fluidization. Fluidization behavior is also dependent on the particle size distribution and mechanical properties of the powder, so it is necessary for these to be considered in simulating a fluidized bed processor. To predict fluidized bed behavior, it was necessary to choose a numerical method capable of accounting not only for the particle-fluid effects (e.g., drag) but also for particle-wall impacts and particle-particle interactions.

Fluidized bed processors of approximate diameters 140mm, 300mm, and 900mm were simulated using a commercial CFD code with a unique model for particle-fluid simulation. For each processor, multiple flow rates were simulated at isothermal conditions for processor loadings between 0.15kg and 45 kg of a powder with a size distribution from 2 to 1500 microns diameter. The simulations modeled both start-up and steady operation of the processors on a full 3D, time-transient basis within a reasonable calculation time (1-2 days).

This study was performed as a “blind test,” with no results shared with the CFD analyst until the simulations were completed. Simulation results were compared with experimental data for dense phase bed height, elutriated particle trajectory, and patterns in the fluidization flow structure. Due to scale-dependent effects, the flow structure varied from a pattern approaching spouted flow to a homogeneously fluidized bed. Observations through the sight glasses were also compared to model predictions. The simulation results compared well with both the laboratory measurements and qualitative observations.

The numerical method and simulation approach were found to successfully predict flow behavior of several fluid bed scales, yielding qualitatively correlated information based solely on material properties, processor loading, inlet air flow rate and processor geometry. This implies that CFD of this kind may be used to assist in determining select operating parameters for pharmaceutical processors and in scaling processes to different sizes to meet production demands.

Wei Chen received his Ph.D from Clarkson University in Mechanical Engineering in 2006. Currently he holds the position of Senior Research Investigator at Bristol-Myers Squibb Co. and was honored with the Chemistry Leadership Award in 2010. His primary area of work is in modeling including mixing (liquid), dissolution (control release et. al. ), powder blending, fluidized bed and pan drying, reaction kinetics, and fermentation based on the CFD and DEM techniques all for pharmaceutical industry processes. Most notably, he developed an elegant mathematical model that predicts the relative deviation of tablet content uniformity for the pan coating process. The model was a key aspect of one of the first Quality by Design New Drug Applications and has been used in several related products which were also approved by the FDA and other health authorities around the globe.

Through Eco-Efficiency Analysis

Abstract:

Many companies have realized the importance of sustainability as a critical business success factor – that what is good for the environment and society can also be good for their financial bottom line. By prioritizing sustainability as a key strategic focus and managing it similar to other parts of the business, such as marketing and sales, leading companies have been able to better identify and manage risks, enhance brand value and corporate reputation, and, more importantly, facilitate clear, measurable value creation throughout the supply chain.

Sustainable development has been defined as the balance of economic success, ecological protection, and social responsibility. To effectively manage sustainability, a company must be able to measure or otherwise quantify sustainabilty in each of those pillars. Eco-efficiency analysis (EEA) harmonizes two of these pillars – the economy and the environment, EEA is a comprehensive, science-based approach that provides information about the relationship between a product’s or technology’s economic benefits and its impacts on the environment along the entire supply chain and throughout all of its lifecycle stages.

BASF has completed more than 400 EEA studies for a diverse range of products, including chemical intermediates, consumer and personal-care products, vitamins, packaging materials, adhesives, and renewable based products. As a strategic tool, EEA provides the necessary data to support internal investment and product portfolio decisions. Just as importantly, it helps customers and other external stakeholders manage the proliferation of eco-confusion by presenting a large amount of complex data in a clear and easily understood manner.
